When Kara (under brainwashing) first met her long-lost cousin, she didn’t like how he was acting and thought his time on earth made him soft. After defeating her cousin Kara decided to bring him to meet Brainiac and show Superman the might of the Kryptonian Empire. However, after getting to know Kal-El, she slowly began to see things from his point of view and later started to overcoming the brainwashing. Their bond grew significantly in a short period, with Kara even sometimes calling her cousin by his Earth name Clark out of respect.
When she discovered the terrible truth about Brainiac's intentions and overcoming her brainwasheing, Kara realized that she put her cousin in terrible danger and vowed to save him. She was utterly horrified when Brainiac took over Superman’s body and was beside herself with grief and guilt over the whole situation. After Superman is freed from Brainiac's control, she admits that he is right about everything and joins him in the fight against her former father figure. When Brainiac tried to take control of Kara again, Kal-El, with the help of Lois and Jimmy, shows her the love and care she needed for a long time, which finally helps Kara to break free of her programming for good. After Brainiac is destroyed, Clark helps her adjust to life on Earth and Kara starts protecting Metropolis alongside her cousin, with the latter now fully embracing her new home.

Kara learned of Kal-El's adoptive parents when she looked through his memories. At first, Kara (under brainwashing) thought all they did was hold Kal-El back; however, after her cousin helped break her free from Brainiac's brainwashing and when she received a warm welcome once she met them in person during her post recovery from the final battle with Brainiac, she became very fond of them.

Jimmy Olsen[]
He is the first person who Kara interacted with on Earth after saving her from being hit by a truck, unaware of her identity as his best friend's cousin, while she didn't know that he is her cousin's best friend. He gave her a tour on Metropolis, showing the wonders of Earth culture, Jimmy also helps her from finding her cousin, as well of learning of Kara's identity as Clark's cousin. Kara shared Jimmy's desire to explore the unknown, as when the two of them went to the roof of the Daily Planet, she expressed her desire to see a world without rules, restrictions or parameters, explore new places and step into the unknown. This is a common ground and interest between the two of them. Jimmy was one of the two people (the other being Superman) that showed Kara and there is more to life than being a warrior and there is also a whole universe outside of the so called "Kryptonian empire".

Kara was also grateful to Jimmy for assisting her to find Clark. Even in her brainwashed state the time they spent when Kara confronted Clark about wanting to meet him, she considered Jimmy a temptation so how their time together meant something to her even in that state. Much later, Clark discovered she had pictures of Jimmy and quickly deduced that she has romantic feelings for him, something that caused her to heavily blush and quickly deny. After a brainwashed Kara went on a rampage, Jimmy held on to her while he and his friends assured her that Kara wasn't alone anymore and that he, Kal-El and Lois were there for her. This long-needed act of compassion was enough to give Kara the strength to free herself of Brainiac's control permanently. After the battle Kara woke up in the Kent house and saw Jimmy downstairs sleeping with a "Welcome to Earth" welcome basket and was touched by the gesture. With Kara now living on Earth, she and Jimmy begin hanging out regularly, bringing warmth, happiness and laughter in each other's lives. Though not technically being a couple at the end of season 2, it was later revealed at SDCC that we will see this relationship develop into a something more throughout season 3.

Lois Lane[]
Kara's relationship with Lois started out very rocky, what with Kara kidnapping Clark and with Lois causing emotional pain to Clark during an argument. However, they were willing to work together to rescue Superman from Brainiac. Kara's view on Lois improved significantly after Lois saved Kal-El from the Black Mercy and after an extremely grateful Kara showed Lois genuine remorse for her past actions under Brainiac's control the two ladies slowly begin to trust and respect each other. When Kara was under the effect of the "Eradicator" protocol, Lois showed that she now actually cared about her by hugging her and reminded her to not let Brainiac win, and it was her turn to let her, Clark and Jimmy help her now. Kara was so moved and empowered by hearing all this, it finally released her from Brainiac's influence at long last. After Brainiac was defeated, Kara and Lois became very good friends.

Kara saw Brainiac as the "light of Kypton" until she learned of his true sinister nature.
Brainiac was Kara's adoptive father, who found and took her in since she was a baby. At some point he placed Kara into intense brainwashing wich made her obedient and blindly followed his commands. Being okay with conquering other worlds beside him as he had brainwashed her to be a bloodthirsty warrior. Though despite being Brainiac's adoptive daughter, Kara considered him to be strict and told Jimmy that, despite feeling guilty, she went ahead to Earth without him to explore Earth without the need for parameters and rules he puts on her. Even so, she is still brainwashed wen being obeyed his commands her to kill her own cousin until Brainiac changed his mind.
Her loyalty to him ended when the real kara learns the terrible truth that Brainiac is a warmongering tyrant willing to kill those who refuse his rule and that he brainwashed her to commit horrible atrocities to the worlds he conquered and the violent actions did on Earth. At first, even after overcoming her brainwashing, she was in denial coming to the idea that Brainiac was malfunctioning and thought she could reason with him; after realizing that wasn't the case at all, and his true intentions and what he really planned to do with Clark, Kara's turns against her adoptive father and reluctantly becomes determined to end his life to stop him from continuing his conquering. Though she is shown to be saddened by this and cried when she assumed she killed him when she turned against him and when Brainiac acknowledged he did indeed love her, bringing her more pain.

After witnessing the devastation Brainiac caused firsthand, she finally saw him for the monster he truly was and Kara renounced him and cuting all ties with him, and even stopped calling him "Father". During the final battle, Brainiac activated the 'Eradicator Protocol' to turn Kara into a weapon against Metropolis, while simultaneously bombarding her with the ideas that without him to serve in obedience, she would be alone, and she was worthless. But, thanks to her cousin and new Earth friends reminding her of her own worth in how she helped save her cousin from Brainiac, that she could be her own person, and that she wasn't alone anymore, Kara was finally able to permanently break free of Brainiac's brainwashing, and consequently unleashed all her fury upon the evil machine. It was in fact Kara herself that dealt the finishing blow to Brainiac, ripping out his CPU and ending his reign of terror, with the rest of his body falling into the sun. When she woke up days later, she looked at the CPU with uncertainty, though keeps it regardless, despite disowning Brainiac as a father.

Kara first encountered Lex Luthor when she and her new friends came to warn of Brainiac's invasion.
Other[]
Brain & Monsieur Mallah[]
Even though she has not interacted with them much Kara was willing to work with them and after returning the earth and their ship broke in two, she shows concern for Brain and Mallah's safety by telling them to get away. Over the next week after the failed invasion, Mallah and the Brain rebuild Cadmus with Kara and Jimmy's help.
Steve Lombard[]
Even though they just met Steve tried to lift Kara's spirit and showed he believed she could be a hero too just like her cousin.
